Sumario: | A method for the preservation of microbial cultures in vacuum is described, modifying SORDELLI`s procedure. The modification consists in the use of a single tube of 7 mm. inner diameter and 1 mm. of wall thickness. The culture is introduced and stuck to the bottom of the sterilized tubes, the cotton plug is pushed and a few pieces of KOH are introduced and compressed between two cotton plugs. The label with the required references is put and the tube is sealed through a previously narrowed place, after a good vacuum is obtained.
The procedure does not modify fundamentally the original method, but has the advantages of the use of a single tube and the guarantee of vacuum permanence. |
