Actually, the natural product use become interesting because its potential chemotherapeutic activity. T. absinthioides (Hook. & Arn.) DC is an aromatic plant currently used because its medicinal and culinary purposes. Its infusions have been used since Aymara and Huarpe traditional medicines because its anti-cholesterolemic, balsamic, expectorants, for the treatment of renal and hepatic insufficiencies, diabetes and ?empacho?. Studies realized in our laboratory have demonstrated the cytotoxic properties of T. absinthioides aqueous extract (TaAE) against human tumoral cell lines. Over no-tumoral cell lines, cytotoxicity was selectively reduced. Performed studies on laboratory animals allowed to demonstrate that TaAE oral administration do not induce acute or repeated dose toxicity (doses up to 2000 mg/kg and 1000 mg/kg/day, respectively), Moreover, the oral administration of TaAE at 300 mg/animal/day induce a significant increases in the median survival of animals with colorectal cancer. Recently, we studied the capability of TaAe to act synergistically with several chemotherapeutics. Carboplatin is one of the agents reported as synergistic. In accordance to the presented information, TaAE is a promising natural compound in the cancer treatment research. In particular, the current project propose to characterize chemically the TaAE, to describe its in vitro and in vivo activity against melanoma and to study the pharmacologic interaction with carboplatin. Dentro de los diferentes tipos de cáncer de piel, específicamente, la mortalidad relacionada al melanoma presenta tasas continuamente en aumento. A pesar de que el melanoma tiene una incidencia menor al 2%, esta patología es la responsable de más del 90% de las muertes asociadas al cáncer de piel. Aunque se han desarrollado distintos enfoques terapéuticos para el tratamiento del melanoma, incluyendo: cirugía, radiación y quimioterapia; la incidencia de la enfermedad continúa en aumento Es por ello, que resulta necesario el desarrollo de nuevas estrategias terapéuticas que presenten mayor eficiencia. El uso de productos naturales (PN) en el tratamiento del cáncer se ha convertido en un gran foco de interés debido a su potencial actividad quimioterápica. T. absinthioides (Hook. & Arn.) DC es una planta aromática comúnmente utilizada por propiedades medicinales y como aditivo culinario en diversas regiones de Chile y Argentina. Sus infusiones han sido empleadas desde los tiempos de la medicina tradicional Aymará y Huarpe por sus propiedades anticolesterolémicas, balsámicas, expectorantes, para el tratamiento de la insuficiencia renal y hepática, la diabetes y el empacho. Estudios realizados en nuestro laboratorio han demostrado las propiedades citotóxicas del extracto acuosos de T. absinthioides (EATa) sobre líneas celulares tumorales humanas; mientras que en líneas celulares humanas no-tumorales, el efecto citotóxico resultó selectivamente disminuido. Experimentos realizados en animales de laboratorio, demostraron que la administración oral del EATa no induce toxicidad aguda, ni a dosis repetidas (dosis hasta 2000 mg/kg y 1000 mg/kg/día, respectivamente). Además, la administración oral del extracto a dosis de 300 mg/animal/día produjo un aumento significativo de la sobrevida en animales con cáncer colorectal inducido mediante la administración de di-metil-hidrazina (DMH). Recientemente, hemos estudiado la capacidad del EATa de actuar sinérgicamente con diferentes quimioterápicos, utilizando líneas celulares de adenocarcinoma mamario (MCF-7). Entre los fármacos reportados como sinérgicos se encuentra carboplatino. De acuerdo a la información presentada, el EATa es un compuesto natural prometedor para la investigación en el tratamiento del cáncer. En particular, el presente proyecto propone caracterizar químicamente el extracto acuoso, describir in vitro e in vivo sus acciones contra el melanoma y estudiar su interacción farmacológica con el carboplatino. |
